Study on microencapsulation of Anthocidaris crassispina gonad oil

摘要: 为开发品质较好的紫海胆(Anthocidaris crassispina)黄油微胶囊,以辛烯基琥珀酸酯化淀粉(HI-CAP100)和β-环糊精为壁材,通过喷雾干燥制备紫海胆黄油微胶囊,以乳化液稳定性及包埋率为指标,通过单因素试验,研究紫海胆黄油微胶囊的壁材比例、乳化液浓度及油脂添加量对微胶囊的影响。以单因素试验的结果为基础,采用正交试验得出紫海胆黄油微胶囊化的最优制备工艺,并检测其理化性质和卫生指标。结果显示,最优制备工艺为:壁材比例4:2(HI-CAP100:β-环糊精),乳化液浓度17%,油脂添加量15%,经过IKA均质乳化机乳化30 min,料理机均质5min,在进风温度180 ℃、出风温度80℃、进样速度500mL/h的条件下进行喷雾干燥时,得到的包埋率和乳化液稳定性分别为95.7%和96.8%,紫海胆黄油微胶囊的水分含量2.82%,溶解度71.94%,自流角22.05°,菌落总数480 CFU/g,大肠菌群数<30 MPN/100g,霉菌和酵母<10 CFU/g;金黄色葡萄球菌、沙门氏菌、志贺氏菌皆未检出。紫海胆黄微胶囊的理化性质及安全卫生指标均符合行业标准。

Abstract: To develop a better quality microencapsulation of Anthocidaris crassispina gonad oil, it was microencapsulated in the wall material of n-octenylsuccinate-derivatized starch(HI-CAP100) and β-cyclodextrin.In this study, spray drying was applied in the preparation of Anthocidaris crassispina gonad oil micro-encapsulation, emulsion stability and embedding rate were used as evaluation indexes.Through single factor experiment to study the effects of wall material ratio, the concentration of oil emulsion and the oil addition on the microcapsules.Based on single factor experimen,take orthogonal method was used to optimize the process conditions,and detected the  physicochemical properties, and hygiene indicators.The results showed that,the optimum conditions of Anthocidaris crassispina gonad oil micro-encapsulation were: the ratio of HI-CAP100 and β-cyclodextrin was 4:2,solids content 17% ,the ratio of oil accounts for wall material 15%.After being emuldified with IKA homogenizer for 30min and homogenizing with a cooking machine for 5 min,the inlet air temperature of spray dryer 180 ℃,outlet air temperature 80 ℃, and the sampling rate of 500mL/h,under which,  embedding rate of 95.7%, emulsion stability of 96.8% were obtained. The Anthocidaris crassispina gonad oil microcapsule in the moisture content was 71.94%,  the solubility was71.94%,the gravity angle was 22.05°,the total number of colonies was 480 CFU/g, the number of coliforms was less than 30 MPN/100g,mold and yeast were less than 10 CFU/ g,Salmonella, Shigella,Staphylococcus aureus were not detected. The physicochemical properties and safety and hygiene indicators of Anthocidaris crassispina gonad oil microcapsules are in line with industry standards.
